Amines as key building blocks in Pd-assisted multicomponent processes

In the last few years, palladium-mediated three-component synthesis has emerged as an important synthetic methodology to gain access to nitrogen-containing structures. The latest developments in this area are discussed in this review.

Stannylated allyl carbonates as versatile building blocks for the diversity oriented synthesis of allylic amines and amides.

Stannylated allylic carbonates are suitable substrates for Pd-catalyzed allylic aminations. In DMF and with [allylPdCl](2) as catalyst the stannylated allyl amines formed can be directly coupled with electrophiles according to the Stille protocol, giving rise to highly functionalized building blocks in excellent yields.

Peptide bond isosteres: ester or (E)-alkene in the backbone of the collagen triple helix.

[structure: see text] Collagen is the most abundant protein in animals. Interstrand N-H...O=C hydrogen bonds between backbone amide groups form a ladder in the middle of the collagen triple helix. Isosteric replacement of the hydrogen-bond-donating amide with an ester or (E)-alkene markedly decreases the conformational stability of the triple helix. Thus, this recurring hydrogen bond is critica...

Synthesis of Functionalized Aryl-vinyl Ethers as Building Blocks and Auxiliaries in Organic Synthesis

5- hydroxyisoquinoline undergoes a smooth reaction with alkyl propiolates in the presence of triphenylphosphine (15 mol%) to produce the corresponding (E)-alkyl 3-(isoquinolin-5-yloxy) acrylate in good yields. When the reaction was performed by 2-methyl 4-hydroxyquinoline and 3-nitro 4-hydroxypyridine, similar aryl vinyl ethers were obtained.
